Rose Gottlieb uses ammonia to break down any lipo protein bonds in milk allowing all the fat to then be dissolved by ether.

Milk is weighed into a tube and a mixture of Industrial Methylated Spirits and ammonia added and mixed. Diethyl ether and Petroleum ether added and the mixture is shaken on a Gerhardt LS500 shaker.

After shaking the layers separate and the ether layer that now contains all the fat removed.

More ethers are added to the original glassware and shaken again. The ether is removed again and added to the first extract.

This is repeated and the combined three extracts are dried and weighed.

The weight is then compared to the starting weight of the sample to determine % Total Fat.
